Duchenne muscular dystrophy (DMD) is a fatal muscle disease caused by the lack of dystrophin, which maintains muscle membrane integrity. We used an adenine base editor (ABE) to modify splice donor sites of the dystrophin gene, causing skipping of a common DMD deletion mutation of exon 51 (∆Ex51) in cardiomyocytes derived from human induced pluripotent stem cells, restoring dystrophin expression. Prime editing was also capable of reframing the dystrophin open reading frame in these cardiomyocytes. Intramuscular injection of ∆Ex51 mice with adeno-associated virus serotype-9 encoding ABE components as a split-intein trans-splicing system allowed gene editing and disease correction in vivo. Our findings demonstrate the effectiveness of nucleotide editing for the correction of diverse DMD mutations with minimal modification of the genome, although improved delivery methods will be required before these strategies can be used to sufficiently edit the genome in patients with DMD.Perovskite solar cells have emerged as one of the most promising thin-film photovoltaic (PV) technologies and have made a strong debut in the PV field. However, they still face difficulties with up-scaling to module-level devices and long-term stability issue. Here, we report the use of a room-temperature nonvolatile Lewis base additive, diphenyl sulfoxide(DPSO), in formamidinium-cesium (FACs) perovskite precursor solution to enhance the nucleation barrier and stabilize the wet precursor film for the scalable fabrication of uniform, large-area FACs perovskite films. With a parallel-interconnected module design, the resultant solar module realized a certified quasi-stabilized efficiency of 16.63% with an active area of 20.77 cm2 The encapsulated modules maintained 97 and 95% of their initial efficiencies after 10,000 and 1187 hours under day/night cycling and 1-sun equivalent white-light light-emitting diode array illumination with maximum power point tracking at 50°C, respectively.During the Mid-Miocene Climatic Optimum [MMCO, ~14 to 17 million years (Ma) ago], global temperatures were similar to predicted temperatures for the coming century. Limited megathermal paleoclimatic and fossil data are known from this period, despite its potential as an analog for future climate conditions. Here, we report a rich middle Miocene rainforest biome, the Zhangpu biota (~14.7 Ma ago), based on material preserved in amber and associated sedimentary rocks from southeastern China. The record shows that the mid-Miocene rainforest reached at least 24.2°N and was more widespread than previously estimated. Our results not only highlight the role of tropical rainforests acting as evolutionary museums for biodiversity at the generic level but also suggest that the MMCO probably strongly shaped the East Asian biota via the northern expansion of the megathermal rainforest biome. This energy-intensive process dictates that metal mining remains among the foremost CO2 emitters and mine waste is the single largest waste form by mass. We propose a new approach, electrokinetic in situ leaching (EK-ISL), and demonstrate its applicability for a Cu-bearing sulfidic porphyry ore. In laboratory-scale experiments, Cu recovery was rapid (up to 57 weight % after 94 days) despite low ore hydraulic conductivity (permeability = 6.1 mD; porosity = 10.6%). Multiphysics numerical model simulations confirm the feasibility of EK-ISL at the field scale. We use quantum electrodynamics to show how the particle's momentum uncertainty determines the optical coherence of the emitted light. We find that the temporal duration of Cherenkov radiation, envisioned for almost a century as a shock wave of light, is limited by underlying entanglement between the particle and light. Our findings enable new capabilities in electron microscopy for measuring quantum correlations of shaped electrons. Last, we propose new Cherenkov detection schemes, whereby measuring spectral photon autocorrelations can unveil the wave function structure of any charged high-energy particle.Geodetic, seismic, and geological evidence indicates that West Antarctica is underlain by low-viscosity shallow mantle. Thus, as marine-based sectors of the West Antarctic Ice Sheet (WAIS) retreated during past interglacials, or will retreat in the future, exposed bedrock will rebound rapidly and flux meltwater out into the open ocean. Previous studies have suggested that this contribution to global mean sea level (GMSL) rise is small and occurs slowly. We challenge this notion using sea level predictions that incorporate both the outflux mechanism and complex three-dimensional viscoelastic mantle structure. In the case of the last interglacial, where the GMSL contribution from WAIS collapse is often cited as ~3 to 4 meters, the outflux mechanism contributes ~1 meter of additional GMSL change within ~1 thousand years of the collapse. Using a projection of future WAIS collapse, we also demonstrate that the outflux can substantially amplify GMSL rise estimates over the next century.
