Severe withdrawal symptoms triggered by cessation of long-term opioid use deter many individuals from seeking treatment. Opioid substitution and α2-adrenergic agonists are the current standard of pharmacotherapy for opioid use disorder in western medicine; however, each is associated with significant complications. Heantos-4 is a non-opioid botanical formulation used to facilitate opioid detoxification in Vietnam. While ongoing clinical use continues to validate its safety and effectiveness, a mechanism of action accounting for these promising effects remains to be specified. Here, we assess the effects of Heantos-4 in a rat model of morphine-dependence and present evidence that alleviation of naloxone-precipitated somatic withdrawal signs is related to an upregulation of mesolimbic dopamine activity and a consequent reversal of a hypodopaminergic state in the nucleus accumbens, a brain region implicated in opioid withdrawal. A central dopaminergic mechanism is further supported by the identification of l-tetrahydropalmatine as a key active ingredient in Heantos-4, which crosses the blood-brain barrier and shows a therapeutic efficacy comparable to its parent formulation in attenuating withdrawal signs. The anti-hypodopaminergic effects of l-tetrahydropalmatine may be related to antagonism of the dopamine autoreceptor, thus constituting a plausible mechanism contributing to the effectiveness of Heantos-4 in facilitating opioid detoxification.The ability to predict others' behaviour represents a crucial mechanism which allows individuals to react faster and more appropriately. To date, several studies have investigated humans' ability to predict conspecifics' behaviour, but little is known on our ability to predict behaviour in other species. Here, we aimed to test humans' ability to predict social behaviour in dogs, macaques and humans, and assess the role played by experience and evolution on the emergence of this ability. For this purpose, we presented participants with short videoclips of real-life social interactions in dog, child and macaque dyads, and then asked them to predict the outcome of the observed interactions (i.e. aggressive, neutral or playful). Participants were selected according to their previous species-specific experience with dogs, children and non-human primates. Our results showed a limited effect of experience on the ability to predict the outcome of social interactions, which was mainly restricted to macaques. Moreover, we found no support to the co-domestication hypothesis, in that participants were not especially skilled at predicting dog behaviour. Finally, aggressive outcomes in dogs were predicted significantly worse than playful or neutral ones. Based on our findings, we suggest possible lines for future research, like the inclusion of other primate species and the assessment of cultural factors on the ability to predict behaviour across species.The tension between selfishness and prosocial behavior is crucial to understanding many social interactions and conflicts. Currently little is known how to promote prosocial behaviors, especially in naturally occurring relationships outside the laboratory. We examined whether a psychological micro-intervention would promote prosocial behaviors in couples. Across two studies, we randomized dyads of couples to a micro-intervention (15 min), which increased prosocial behaviors by 28% and decreased selfish behaviors by 35% a week later in behavioral games in a dose-response manner. Using event sampling methodology, we further observed an increase in prosocial behaviors across one week that was most pronounced in participants who received the intervention. These results from the laboratory and everyday life are important for researchers interested in prosocial behavior and selfishness and have practical relevance for group interactions.With global warming, the carbon pool in the degradation zone of permafrost around the Arctic will gradually be disturbed and may enter the atmosphere in the form of released methane gas, becoming an important factor of environmental change in permafrost areas. We selected the northwestern section of the Xiao Xing'an Mountains in China as the study area, located in the degradation zone on the southern margin of the permafrost region in Eurasia, and set up multiple study monitoring areas equipped with methane concentration sensors, air temperature sensors, pore water pressure sensors and soil temperature sensors for long-term monitoring of data changes using the high-density electrical method, ground penetrating radar and on-site drilling to survey the distribution of frozen soil and geological conditions in the study area, combined with remote sensing images of Sentinel-2 L1C and unmanned aerial vehicle photographs and three-dimensional image reconstruction, analysis of fire activities and related geological elue, and the surface methane concentration reaches an annual high value. The high concentration of methane gas entering the surface in this stage is expected to increase the risk of wildfire in the permafrost degradation area in two ways (increasing the regional air temperature and self-combustion), which may be an important factor that leads to a seasonal wildfire frequency difference in the permafrost zone of Northeast China and Southeast Siberia, with the peak in spring and autumn and the monthly maximum in spring. The increase in the frequency of wildfires is projected to further generate positive feedback on climate change by affecting soil microorganisms and soil structure. Southeastern Siberia and northeastern China, which are on the southern boundary of the permafrost region of Eurasia, need to be targeted to establish fire warning and management mechanisms to effectively reduce the risk of wildfires.Parkinson's disease (PD) is a neurodegenerative disease characterized by the loss of dopaminergic neurons in the substantia nigra resulting in severe and progressive motor impairments. However, the mechanisms underlying this neuronal loss remain largely unknown. PF2545920 Oxidative stress and ER stress have been implicated in PD and these factors are known to activate the integrated stress response (ISR). Activating transcription factor 4 (ATF4), a key mediator of the ISR, and has been reported to induce the expression of genes involved in cellular homeostasis. However, during prolonged activation ATF4 can also induce the expression of pro-death target genes. Therefore, in the present study, we investigated the role of ATF4 in neuronal cell death in models of PD. We demonstrate that PD neurotoxins (MPP+ and 6-OHDA) and α-synuclein aggregation induced by pre-formed human alpha-synuclein fibrils (PFFs) cause sustained upregulation of ATF4 expression in mouse cortical and mesencephalic dopaminergic neurons. Furthermore, we demonstrate that PD neurotoxins induce the expression of the pro-apoptotic factors Chop, Trb3, and Puma in dopaminergic neurons in an ATF4-dependent manner.
