[BMB Reports 2021; 54(6) 323-328].Cancer stem cells (CSCs) are a subpopulation of cancer that can self-renew and differentiate into large tumor masses. Evidence accumulated to date shows that CSCs affect tumor proliferation, recurrence, and resistance to chemotherapy. Recent studies have shown that, like stem cells, CSCs maintain cells with self-renewal capacity by means of asymmetric division and promote cell proliferation by means of symmetric division. This cell division is regulated by fate determinants, such as the NUMB protein, which recently has also been confirmed as a tumor suppressor. Loss of NUMB expression leads to uncontrolled proliferation and amplification of the CSC pool, which promotes the Notch signaling pathway and reduces the expression of the p53 protein. NUMB genes are alternatively spliced to produce six functionally distinct isoforms. An interesting recent discovery is that the protein NUMB isoform produced by alternative splicing of NUMB plays an important role in promoting carcinogenesis. In this review, we summarize the known functions of NUMB and NUMB isoforms related to the proliferation and generation of CSCs. However, the biological effects of human collagen α-1 type I (hCOL1A1) on various skin properties have not been investigated. Here, we isolate and identify the hCOL1A1-collagen effective domain (CED) which promotes collagen type I synthesis. Recombinant hCOL1A1-CED effectively induces cell proliferation and collagen biosynthesis in HDFs, as well as increased cell migration and elastin production. Based on these results, hCOL1A1-CED may be explored further for its potential use as a preventative agent against skin aging. The post-translational modification of LSD1 is critical for the regulation of many of its biological processes. Phosphorylation of serine 112 of LSD1 by protein kinase C alpha (PKCα) is crucial for regulating inflammation, but its physiological significance is not fully understood. This study aimed to investigate the role of Lsd1-S112A, a phosphorylation defective mutant, in the cigarette smoke extract/LPS-induced chronic obstructive pulmonary disease (COPD) model using Lsd1SA/SA mice and to explore the potential mechanism underpinning the development of COPD. We found that Lsd1SA/SA mice exhibited increased susceptibility to CSE/LPS-induced COPD, including high inflammatory cell influx into the bronchoalveolar lavage fluid and airspace enlargement. Additionally, the high gene expression associated with the inflammatory response and oxidative stress was observed in cells and mice containing Lsd1-S112A. Similar results were obtained from the mouse embryonic fibroblasts exposed to a PKCα inhibitor, Go6976. Thus, the lack of LSD1 phosphorylation exacerbates CSE/LPS-induced COPD by elevating inflammation and oxidative stress.This paper describes research about a historical bottle found in the Polish town of Skarszewy in 2004. Upon discovery, the find was labeled "In Nazareth Aechter Jerusalemer Balsam im goldnen Engel", sealed and ⅓ filled with liquid. The Jerusalem Balsam mentioned on the label was a popular medicament in Europe in the 18th century. From 1719 it was produced by Father Antonio Menzani da Cuna in the Franciscan Pharmacy at the convent of Saint Savior in Jerusalem. In the 19th century, the Balsam became extremely popular in Silesia thanks to the hermit Johannes Treutler from Mariańska Hill near Kłodzko. It's fame spread north to Prussia and south to Bohemia (Czechia). After the hermit's death, the license for production was obtained by the owner of the Mohren-Apotheke pharmacy, but he had to deal with unfair competition from other pharmacies counterfeiting the Balsam. An attempt was made to determine where the found bottle came from. In the course of the research, it was found that the medicine certainly does not come from authorized production sources, as evidenced by accurate label comparisons.A high proportion of hospitalizations is attributable to the prevalence of adverse drug events. This retrospective study included outpatients and inpatients to determine the prevalence of adverse drug events and if polypharmacy increases it. The prevalence, classification, and causality of adverse drug events were assessed based on medical records, laboratory values, and other data. Multivariate analysis (multiple logistic regression analysis) was performed with the presence or absence of adverse drug events at the time of the visit as the dependent variable and items for which the P-value was less then 0.25 in the univariate analysis as independent variables. The prevalence of adverse drug events was 13.0%, 10.9%, and 16.0% among all patients, the outpatient group, and the inpatient group, respectively. Multivariate analysis showed that polypharmacy (≥5 drugs) significantly increased the risk of adverse drug events in all patients. The prevalence of adverse drug events significantly increased with each additional drug used. We expect that minimizing the number of medications through moderation of the number of prescription drugs and elimination of polypharmacy will reduce the number of outpatient visits and hospitalizations due to adverse drug events.The objective of this study was to determine the number of patients on the national level that took macrolide antibiotics along with chronic statin therapy in Croatia in the period from 2002 to 2015, and to analyse prescription patterns. In 2002, statins were used in the treatment of 2.6% of the total number of insured persons in Croatia. By 2015, this number increased to 8.4%. ZEN-3694 inhibitor In the period studied, on average 15.3% of the patients on statin therapy were co-prescribed macrolide antibiotics. Erythromycin was combined with different statins on average in 1.4% of cases, clarithromycin in 25.5% and azithromycin in 73.2% of the cases. Relative frequency of combining statins with macrolides was similar for all statins. On average, 11.5% of patients on concomitant statin-macrolide therapy were taking high-dose statins. On average, 90% of these co-prescriptions can lead to potentially clinically significant DDIs (X, D, C). The co-prescription of statins and macrolide antibiotics in the Republic of Croatia is increasing.
