We apply a mixed-payer economy model to study the effects of changes in the generosity of children's public health insurance programs - measured by Medicaid and Children's Health Insurance Program income thresholds - on substance use disorder (SUD) treatment provider behavior. Using government data on specialty SUD treatment providers over the period 1997-2011 combined with a two-way fixed-effects model and local event study, we show that increases in the generosity of children's public health insurance induce providers to participate in some, but not all, public markets. Our effects appear to be driven by non-profit and government providers. Non-profit providers also appear to increase treatment quantity slightly in response to coverage expansions.This paper investigates the role of biased health perceptions as a potential driving force of risky health behaviors. In this study, mRNA expressions of MRC1 gene in nine porcine organs/tissues were compared between Laiwu (LW) and Yorkshire × Landrace crossbred (YL) pigs prior to and post PCV2 infection. We found that, for pigs uninfected with PCV2, MRC1 mRNA expressions in the lung, spleen, large intestine, small intestine and mesenteric lymph node tissues of LW were significantly higher than those of YL pigs (P less then 0.05). After PCV2 infection, MRC1 mRNA levels in the liver, kidney and mesenteric lymph node were significantly increased in LW pigs (P less then 0.05); while, significantly decreased in the heart and lung tissues of YL pigs (P less then 0.05). The transcriptional activity of porcine MRC1 promoter was further analyzed to investigate the molecular mechanism underlying these expressional differences in response to PCV2 infection. Luciferase assay indicated that a 14 bp indel polymorphism "GTTTTTTTTTTTTT" at the site -864 of MRC1 promoter contributed to the transcriptional activity. The frequency of 14 bp insertion in LW and Dapulian pigs, generally resistant to PCV2 infection, was higher than that in Duroc, Landrace and Yorkshire pigs, which were sensitive to PCV2 infection. The promoter with 14 bp insertion displayed higher MRC1 transcription level both prior to and post PCV2 infection compared with that carrying no insertion in PK15 cells (P less then 0.01). The results suggest that this 14 bp indel polymorphism is associated with different responses to PCV2 infection by regulating MRC1 transcription.Spills of diluted bitumen (dilbit) from pipelines pose a risk to the health of aquatic organisms, including fish, and with expected increases in production and transportation of dilbit, these risks could increase. To date, the majority of studies have investigated effects of fresh dilbit on aquatic organisms, but little is known about effects of weathered sediment-bound dilbit, including mechanisms of toxicity. The goal of this study was to use 1H NMR based metabolomics to identify altered metabolites and pathways in early life-stages of zebrafish (Danio rerio) exposed to a sediment derived water-soluble fraction of dilbit (SDWSF) to better understand mechanisms of adverse effects. Zebrafish embryos exposed to the SDWSF until 120 h post-fertilization exhibited increased prevalence of pericardial edema, yolk sac edema, and swim bladder malformations that are typical of exposure to fresh dilbit. Concentrations of nine metabolites (alanine, glutamine, lysine, threonine, tyrosine, betaine, taurine, inosine, and glycerol) were significantly altered in embryos exposed to SDWSF. Pathway topology analysis revealed four potentially impacted pathways 1) phenylalanine, tyrosine, and tryptophan biosynthesis, 2) taurine and hypotaurine metabolism, 3) alanine, aspartate, and glutamate metabolism, and 4) glycine, serine, and threonine metabolism. Altered metabolites were linked to several biological process, that when perturbed could be key events in mechanisms of developmental effects observed in embryos. Future studies should further investigate the role of perturbations to these metabolites and pathways to determine the specific role they might play in adverse effects of exposure to dilbit.Accurate segmentation of cerebral vasculature and a quantitative assessment of its morphology is critical to various diagnostic and therapeutic purposes and is pertinent to studying brain health and disease. However, this is still a challenging task due to the complexity of the vascular imaging data. We propose an automated method for cerebral vascular segmentation without the need of any manual intervention as well as a method to skeletonize the binary segmented map to extract vascular geometric features and characterize vessel structure. We combine a Hessian-based probabilistic vessel-enhancing filtering with an active-contour-based technique to segment magnetic resonance and computed tomography angiograms (MRA and CTA) and subsequently extract the vessel centerlines and diameters to calculate the geometrical properties of the vasculature. Our method was validated using a 3D phantom of the Circle-of-Willis region, demonstrating 84% mean Dice similarity coefficient (DSC) and 85% mean Pearson's correlation coal length (3.46 ± 0.99 m vs. 2.20 ± 0.67 m for CTA), lower total volume (61.80 ± 18.79 ml vs. 34.43 ± 22.9 ml for CTA), lower average diameter (2.4 ± 0.21 mm vs. 2.18 ± 0.07 mm for CTA), and lower average branch length (4.81 ± 1.97 mm vs. 8.68 ± 2.03 mm for MRA), respectively. We additionally studied the change in vascular features with respect to aging and imaging modality. While we observed differences between features as a result of aging, statistical analysis did not show any significant differences, whereas we found that the number of branches were significantly different (p less then 0.05) between the two imaging modalities (201 ± 73 for MRA vs. 189 ± 69 for CTA). Our segmentation and feature extraction algorithm can be applied on any imaging modality and can be used in the future to automatically obtain the 3D segmented vasculature for diagnosis and treatment planning as well as to study morphological changes due to stroke and other cerebrovascular diseases (CVD) in the clinic.
