An analytical method was developed and validated for the determination of three polyether ionophores (monensin, lasalocid, and salinomycin) in 60 samples of Brazilian Minas Frescal cheese by UHPLC-MS/MS. Linearity ranged from 1 to 8 μg kg-1 for monensin and salinomycin, and from 0.50 to 4 μg kg-1 for lasalocid. Limits of detection and quantitation were 0.50 μg kg-1 and 1 μg kg-1, respectively, for both monensin and salinomycin, and 0.25 μg kg-1 and 0.50 μg kg-1, respectively, for lasalocid. Recoveries were between 69% and 84% with coefficients of variation up to 16.28% for repeatability and 13.79% for intermediate precision. A total of 60 samples of Minas Frescal cheese were analysed and only monensin residues were found. Monensin was detected in 55% of the samples and quantified in 5 of them at mean levels varying from 1.00 to 1.73 μg kg-1. The proposed method demonstrated the suitability for monitoring these substances in cheese.Leishmaniasis, a category 1 disease, has remained neglected for decades, and therefore, has developed into a severe health problem worldwide. Unfortunately, the available antileishmanial drugs are limited, and the parasites have shown an inevitable resistance toward most of these drugs. All these factors pose a barrier to control the parasite at present. Hence, new strategies are needed to develop more effective and less toxic nanomedicines that could treat and manage the Leishmania parasite. One of these effective strategies is to construct nanometals with biologically active molecules that could possess dynamic antileishmanial activities with desirable biocompatibility. In this review paper, antileishmanial potencies of different metal nanoparticles, with particular emphasis on biogenic metal nanoparticles from 2011 to 2019, are summarized. The mechanisms by which metal-based nanomedicines kill Leishmania are also discussed.Ocular diseases are an important category in equine medicine; however, most articles regarding histologic ocular lesions in horses are exclusive to a specific condition and do not provide a complete review of clinically significant ocular disease frequency in a diagnostic laboratory. We reviewed sections of equine eyes from 140 cases (98 enucleations [biopsies] and 42 autopsies) with clinically relevant ocular alterations at 2 diagnostic centers in the United States. The most common primary conditions were non-traumatic keratitis (36), equine recurrent uveitis (ERU; 31), traumatic injuries (22), ocular and periocular neoplasms (19), and uveitis and/or endophthalmitis resulting from sepsis (18). Congenital anomalies (3) and retinal atrophy and detachment alone (3) were infrequent. Non-traumatic keratitis was frequently accompanied by anterior uveitis (22), corneal rupture (16), pre-iridal fibrovascular membrane formation (13), and secondary mycotic infection (11). Based on a VSV pseudovirus production system, a pseudovirus-based neutralization assay has been developed for evaluating neutralizing antibodies against SARS-CoV-2 in biosafety level 2 facilities. The key parameters for this assay were optimized, including cell types, cell numbers, virus inoculum. When tested against the SARS-CoV-2 pseudovirus, SARS-CoV-2 convalescent patient sera showed high neutralizing potency, which underscore its potential as therapeutics. The limit of detection for this assay was determined as 22.1 and 43.2 for human and mouse serum samples respectively using a panel of 120 negative samples. The cutoff values were set as 30 and 50 for human and mouse serum samples, respectively. This assay showed relatively low coefficient of variations with 15.9% and 16.2% for the intra- and inter-assay analyses respectively. Taken together, we established a robust pseudovirus-based neutralization assay for SARS-CoV-2 and are glad to share pseudoviruses and related protocols with the developers of vaccines or therapeutics to fight against this lethal virus.Aim Therapeutically targeting cancer stem cells (CSCs), which play a role in tumor initiation and relapse, remains challenging. Materials & methods Novel-formulated platinum nanoparticles (Pt-NPs) supported on polybenzimidazole (PBI)-functionalized polymers and multiwalled carbon nanotubes (MWCNT) were prepared and their effect on CSCs was evaluated. Results Pt-NPs showed homogenous distribution on the surface of MWCNT/PBI composites, with very narrow particle size. MWCNT/PBI/Pt-NPs resulted in a dramatic decrease in the proliferation rate of CSCs but not bone marrow mesenchymal stem cells (BM-MSCs). Quantitative gene expression analysis revealed that MWCNT/PBI/Pt had a significant inhibitory effect on the epithelial-mesenchymal transition and cell cycle markers of CSCs. Conclusion MWCNT/PBI/Pt exhibited a specific cytotoxic effect on breast CSCs but not on adult stem cells.Large population studies estimated that the frequency of food allergies is increasing worldwide. In the last two decades, a 'second wave' of the allergy epidemic has emerged as a growing public health problem. EU regulation strengthened information to consumers about allergens in pre-packed food, since December 2014 it has been extended to non-prepacked foods by the Regulation (EU) No 1169/2011 of the European Commission. The present work aims to present a five-year survey on the presence of nine types of allergen in several foods, including food of animal origin and baby food. The concentration found for each irregular allergen is generally low, but some samples with no gluten indication contained a concentration above 10 mg kg-1 with the highest value of 138.5 mg kg-1 in a dietetic food, during the screening in 2017. These data underline the importance and the necessity of a complete food labelling to protect the majority of allergic individuals.Aim Magnetic particle imaging (MPI) is highly promising for biomedical applications, but optimal tracers for MPI, namely superparamagnetic iron oxide-based contrast agents, are still lacking. Materials & methods The encapsulation of commercially available nanoparticles, specifically synomag®-D and perimag®, into human red blood cells (RBCs) was performed by a hypotonic dialysis and isotonic resealing procedure. The amounts of superparamagnetic iron oxide incorporated into RBCs were determined by Fe quantification using nuclear magnetic resonance and magnetic particle spectroscopy. Results Perimag-COOH nanoparticles were identified as the best nanomaterial for encapsulation in RBCs. Perimag-COOH-loaded RBCs proved to be viable cells showing a good magnetic particle spectroscopy performance, while the magnetic signal of synomag-D-COOH-loaded RBCs dropped sharply. Conclusion Perimag-COOH-loaded RBCs could be a potential tool for MPI diagnostic applications.There is scarce data on the mycotoxin profile in retailed fruit juices in Nigeria. Thirty-five industrially-processed fruit juice samples randomly purchased from retailers in Ogun state, Nigeria, were analysed for the presence of > 650 toxic fungal and plant metabolites using a liquid chromatography tandem mass spectrometric method. Only 18 metabolites, including 3-nitropropionic acid, alternariol methylether and emodin, but excluding citrinin, fumonisin B2, ochratoxin A and patulin, were detected in trace levels in at least one juice sample. Amygdalin, a plant cyanogen, was quantified (2.05-359 µg/L) in 40% of the samples. Although the levels of mycotoxins and toxic plant metabolites found in the juice may be relatively low, daily consumption of juices containing such low levels may contribute to dietary exposures to these natural chemical contaminants in consumers. Fruit juice processors should be encouraged to adhere strictly to good manufacturing practices in order to keep mycotoxins away from the final products.The 2 predominant circulating subtypes of influenza A virus in the dog population, equine-origin H3N8 and avian-origin H3N2, constitute a potential zoonotic risk. We determined the prevalence of influenza A antibodies in 496 dogs in Poland and found 2.21% of sera positive by commercial ELISA. Hemagglutination inhibition (HI) assays indicated 7.25% of sera positive using equine H3N8, swine H3N2, and pandemic H1N1 antigens, with the most frequently detected immune response being to H3N2. Considering interspecies transfer, reassortment ability, and close contact between dogs and humans, infections of dogs with influenza A virus should be monitored.Consideration of cumulative dietary risks is a requirement in the European legislation for setting maximum residue levels for pesticides. Current cumulative exposure assessment methodologies strongly rely on representative occurrence data from food monitoring programmes. This study provides a sensitivity analysis, utilising (i) European consumption data expressed as raw agricultural commodity (RAC) equivalents from 23 different countries as published by the European Food Safety Authority and (ii) all maximum residue levels established for pesticides under European Regulation (EC) No 396/2005. Based on two different degrees of conservatism, market baskets consisting of 16 or 41 RACs, respectively, were identified, covering the majority of the total chronic and acute daily exposure. The coverage of the exposure by these market baskets was tested by comparison of cumulative probabilistic exposure assessments for the German population using all food commodities and those using the reduced sets. It was demonstrated that ≥85% of the total chronic exposure is already covered by 16 RACs, while 41 RACs are required to reach a similarly satisfying coverage of the total acute exposure. Results from this study support resource efficient modelling of complex cumulative assessment scenarios and may help to improve the design of food monitoring programmes with respect to a more efficient assessment of potential consumer risks.Human rights are foundational to the health and well-being of all individuals and have remained a central tenet of nursing's ethical framework throughout history. The purpose of this study is to explore continuity and changes to human rights in nursing codes of ethics in the Canadian context. This study examines nursing codes of ethics between the years 1953 and 2017, which spans the very first code in Canada to the most recently adopted. The historical method is used to compare and contrast human rights language, positioning and descriptions between different code editions. The findings suggest there has been very little change in how human rights have been included within the Canadian nursing codes of ethics. Furthermore, we consider how changes within the nursing profession have influenced the authority of codes of ethics and their ability to support nurses in carrying out ethical obligations specific to human rights. Finally, the impacts and implications of these changes are discussed concerning the protection of human rights in today's healthcare landscape in Canada.
